In the Pacific oyster, Crassostrea gigas, the energy necessary for general metabolism and gametogenesis is mainly provided by the glycogen stored in the vesicular cells. The first level of glycogen metabolism control is the entrance of glucose into cells. The monitoring of the uptake capacity of [U-14C] D-glucose revealed the existence of a saturable component with kinetic constants: Km = 0.71 mM ; Vmax = 0.22 nmol/h/106cells. The use of inhibitors has revealed the presence of carriers belonged to GLuT and SGLT types. A follow-up showed a seasonal variation of the uptake capacity and incorporation of glucose into glycogen, and a high expression of SGLT during mobilization of glycogen and gametes maturation. Collections of frozen tissue samples stand as keystone sources of molecular information to construct biodiversity knowledge, and are particularly challenged if they focus on megadiverse countries. In 1998 the Humboldt Institute (Instituto de Investigación de Recursos Biológicos Alexander von Humboldt) began a tissue collection of Colombian biodiversity (IAvH-CT) and the aim of this work is to present a diagnostic and an historical perspective for that collection, constructed by compiling information and experiences on its management as well as by organizing and curating the information of each catalogued sample. Cryopreservation of Cyrtopodium hatschbachii Pabst (Orchidaceae) immature seeds by encapsulation-dehydration Biocell

The aim of the present study was to investigate the efficiency of the encapsulation-dehydration technique for cryopreservation of Cyrtopodium hastchbachii Pabst seeds. Immature seeds of this species were cryopreserved by an encapsulation-dehydration technique. Seeds of five immature pods, 120 days after pollination, were encapsulated in 3% calcium alginate matrix and pretreated in liquid medium supplemented with 0.08 M sucrose (24 h), 0.15 M sucrose (24 h), 0.25 M sucrose (48 h), 0.5 M sucrose (24 h) and 0.75 M sucrose (24 h) in shaker at 60 rpm. Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied....

Essai de cryoconservation des gamétophytes de l'algue alimentaire Undaria pinnatifida (Laminariale) ArchiMer

Low temperature tolerance was determined for gametophytes of food alga Undaria pinnatifida according to various cryopreservation methods. Survival durations after storage in a medium containing 28% glycerol at −80 °C or in a 5-10% glycerol solution at −196 °C were only 9 and 4 days respectively. On the other hand, cooling at 5 °C/min. and storage at −15 °C for 15 minutes, without cryoprotective medium, did not reduce viability but this was highly decreased by storage for 15 minutes at −30 or −196 °C, with or without DMSO. The morphological cooling injuries were depigmentation and/or a contraction of the cells.

In order to improve oyster culture, many research programs try and propose new methods aiming at improving genetically the product performances and quality (selection, polyploidisation). As for terrestrial animal production, the continuity of the different works as well as the spreading of the genetical progress would be made easier by the development of conservation techniques of the selected genotypes. Cryopreservation, which is often used in selection programs of terrestrial animals or fish farming species, is not always available for bivalves.
